Precision Antibody can help you generate recombinant antibodies. Recombinant antibodies are produced for many reasons such as:

  • Planning for a long-term production of monoclonal antibodies.
  • Hybridoma producing the antibody of interest is a low producer.
  • A small amount of purified antibody is available but lost the hybridoma.
  • Isolated antibody of interest from a phage library or B cells.
  • Need to produce humanized antibody.
  • Need cell line development for fully human or humanized monoclonal antibody.

can help you clone and generate expression vector to be used for recombinant antibody production in CHO and HEK293 cells.

For fully human monoclonal antibody producing hybridoma cells, we can sequence the variable region of H and L chains (VH & VL) using MiSeq.

For mouse and rat monoclonal antibodies, we can amplify the variable region gene by PCR using redundant primer sets.

Optimized H and L chain genes are synthesized and incorporated into appropriate expression vector system. A small amount of recombinant antibody is produced and characterized to confirm their specificity and biophysical characteristics (KD, ka, & kd).

General Process:

  • Sequence analyses of VH & VL
  • Optimize the VH & VL
  • VH gene syntheses.
  • Cloning of VH into IgG vector and sequence confirmation.
  • L chain gene syntheses, cloning into expression vector and sequence confirmation.
  • Transient production of small amounts of IgG.
  • EIA and Octet based confirmation of antigen specificity and binding parameters (KD, ka, & kd).
  • Ultra-low endotoxin plasmid production/purification.
  • Transient IgG production in CHO or HEK293 cells or stable cell line development.
