Precision Antibody is a worldwide recognized leader in developing anti-idiotypic antibodies (anti-idiotype, or anti-Id). Anti-idiotypes are raised against the variable regions of the heavy and light chains of the drug antibody. Anti-idiotypic antibodies can specifically detect the drug antibodies out of all circulating antibodies thus are used in the Pharmacokinetic (PK) Studies and assay for the detection of Anti-Drug Antibody (ADA).
To accommodate requirements for PK assays and ADA assays, Precision Antibody offers the development of mouse or rat anti-Id monoclonal antibodies as well as the development of polyclonal anti-Id antibodies in rat, rabbit, or other species.
Anti-idiotypic antibodies are classified into 2 categories: Antibodies against the paratope and against the idiotope. Drug antibodies against soluble therapeutic targets require the use of anti-idiotope antibodies for PK studies. Both anti-paratope and anti-idiotope antibodies can be used for PK studies of drug antibodies against cell surface targets.
Our team of immunologists has developed a successful immunization protocol that has vastly improved the speed of generations of anti-idiotypic antibodies and its repertoire so that finding anti-idiotype antibodies against distinct epitopes such as idiotopes and paratopes with the same immunization is highly successful.

Upon discussion of strategy with the customer, a typical anti-idiotype antibody development project takes about 60-70 days from start to finish and includes:
- Immunization
- Screening for anti-idiotope and anti-paratope antibodies,
- Screening for antibodies void of human and monkey serum (plasma) interference by either ELISA or Octet.
- Small scale antibody production and purification
- Epitope binning assays to select pairs of antibodies that can be used in sandwich EIA.
- Octet based scouting assay to select high affinity, low off-rate antibodies suited to be used in the sandwich EIA assay.
- Sandwich ELISA assay development to select the pair with desired sensitivity and dynamic range.
Our projects are milestone-based, and customers will get information at each milestone. We provide phase-in pricing to reduce the risk for our clients.
